Optimize Homogenization Protocols
Optimizing Lysing Protocol is critical for efficient homogenization and improve extraction efficiency. The following factors are important to quickly optimize a homogenization protocol:
-Ratio of sample: buffer: beads And leave sufficient airspace in the lysing tube. Overfill will result low homogenization efficiency, while dry grinding may increase the risk of breaking lysing tube caps.
-Inspect samples particles visually for any large pieces and under microscope for evenly distributed particle sizes. The ideal particles size should be at uM ranges.
-Homogenization tube breaking risk: to prevent sample loss from tube breaking, run a protocol with 10% higher speed, evaluate the tube conditions at the end of run. Pay special attention to the caps, bottom of the tubes. If there is clear signs of breakage, dial down the speed or shorter the time.
